UCT product used to explore routes of pain medication administration in ferrets
The Problem
Ferrets are common companion and laboratory animals, but options for treating moderate to severe pain without injection are limited. Understanding whether oral transmucosal (OTM) delivery works for buprenorphine could change how veterinary practitioners manage ferret pain, but first you need solid pharmacokinetic data. That means reliable extraction from plasma.

The Study
Researchers at the University of Georgia ran a pharmacokinetic comparison of intramuscular (IM) vs. oral transmucosal buprenorphine in eight female ferrets. Each animal received 0.04 mg/mL buprenorphine via both routes (crossover design, 10-day washout). Blood was collected at timed intervals up to 720 minutes post-dose through surgically implanted vascular access buttons. Ferrets were monitored for side effects and assigned sedation scores throughout.
The Extraction
Plasma samples (0.1 mL) were diluted in 0.1 M phosphate buffer (pH 6) and extracted by SPE using:

Analysis was performed by LC-MS/MS on a column equivalent to the Selectra® C18, 100 × 2.1 mm, 3 µm (P/N SLC-18100ID21-3UM).
Unendcapped C18 gives you the polar selectivity needed for basic analytes like buprenorphine in biological matrices, without the retention losses you get from fully endcapped phases. Straightforward conditioning, predictable recoveries, clean extracts.
Key Findings
IM pharmacokinetics were consistent with previous literature. OTM administration showed a very short terminal half-life (under one hour), and accurate Cmax/Tmax reporting was limited by proximity of the sampling site to the administration site. Both routes produced comparable sedation effects despite different PK profiles. Further research is needed to fully characterise OTM buprenorphine pharmacokinetics in ferrets.
